ABSTRACT

Cisplatin is a widely used chemotherapeutic drug, but its side effects are a major limiting factor. Nephrotoxicity occurs in one third of patients undergoing cisplatin treatment. The acute tubular injury caused by cisplatin often leads to a defective repair process, which translates into chronic renal disorders. In this way, cisplatin affects tubular cells, and maladaptive tubules regeneration will ultimately result in tubulointerstitial fibrosis. Kinins are well known for being important peptides in the regulation of inflammatory stimuli, and kinin B1 receptor deficiency and antagonism have been shown to be beneficial against acute cisplatin nephrotoxicity. This study aimed to analyze the effects of kinin B1 receptor deletion and antagonism against repeated cisplatin-induced chronic renal dysfunction and fibrosis. Both the deletion and the antagonism of B1 receptor exacerbated cisplatin-induced chronic renal dysfunction. Moreover, the inhibition of B1 receptor increased tubular injury and tubulointerstitial fibrosis after repeated treatment with cisplatin. The balance between M1/M2 macrophage polarization plays an important role in renal fibrosis. Kinin B1 receptor antagonism had no impact on M1 markers when compared to cisplatin. However, YM1, an M2 marker and an important molecule for the wound healing process, was decreased in mice treated with kinin B1 receptor antagonist, compared to cisplatin alone. Endothelin-1 levels were also increased in mice with B1 receptor inhibition. This study showed that kinin B1 receptor inhibition exacerbated cisplatin-induced chronic renal dysfunction and fibrosis, associated with reduced YM1 M2 marker expression, thus possibly affecting the wound healing process.

ABSTRACT

Objetivou-se avaliar a destoxificação da torta de mamona bruta (TMB), por meio de dois produtos alcalinos em diferentes concentrações, e seus efeitos sobre a composição química, a degradabilidade in situ da MS e o fracionamento de proteínas. Utilizou-se o hidróxido de cálcio [Ca(OH)2] e o hidróxido de sódio (NaOH) em duas concentrações (60 e 90 gramas), diluídos em quatro quantidades de água (1.000; 1.500; 2.000 e 2.500mL de água por quilo de TMB). Observou-se que, das diferentes concentrações utilizadas, somente a utilização de 90 e 60 gramas de Ca(OH)2 e NaOH, respectivamente, conseguiu destoxificar 100% da TMB, ambas diluídas em 2.000mL de água. Por outro lado, ao avaliar o tempo mínimo de contato dos reagentes com a TMB para uma máxima destoxificação, observou-se que três horas de contato é o tempo necessário para os reagentes diminuírem em 100% as proteínas citotóxicas, além de não deixar atividade hemaglutinante nesse material. A destoxificação com o NaOH proporcionou maior degradação das proteínas solúveis e da matéria seca, favorecendo a disponibilização do nitrogênio não proteico, estando sua aplicação em escala industrial na dependência de estudos sobre viabilidade operacional e econômica.(AU)


This study aimed to evaluate the detoxification of crude castor (DCC) through two alkaline products in different concentrations and their effects on the chemical composition, in situ degradability of DM and the fractionation of proteins. We used the calcium hydroxide [Ca(OH)2] and sodium hydroxide (NaOH) in two concentrations (60 and 90 grams) diluted in 4 quantities of water (1,000; 1,500; 2,000 and 2,500ml of water per kilo of DCC). It was observed that in the different concentrations used, only the use of 90 and 60 grams of Ca(OH)2 and NaOH, respectively managed to detoxify 100% of the DCC, both diluted in 2,000ml of water. On the other hand, when assessing the minimum time of contact of the reagents with the DCC for maximum detoxification, it was observed that with three hours of contact is the time required for the reagents decrease in 100% of the cytotoxic proteins, in addition to not leave haemagglutinating activity in this material. The detoxification with NaOH provided greater degradation of soluble proteins and degradation of dry matter, favoring the provision of non-protein nitrogen, while its application on an industrial scale is in the dependence of studies on operational feasibility and cost.(AU)

ABSTRACT

This study aimed to evaluate the effect of isolated or combined citric and benzoic acids added to the diets of broiler chickens on performance, allometry of the digestive system organs, intestinal pH and quantity of microorganisms in the jejunum. A total of 840 male Cobb broiler chicks were utilized, distributed in a complete random design in 2 × 2 factorial scheme, supplemented or not with citric acid, and sodium benzoate, with seven replications. At 14 days old, 1mL of a solution containing 1 × 105 sporulated oocysts of Eimeria acervulina per bird was inoculated orally. There was no effect of the acids on the broiler's performance in the 1 to 21-day period. In the total period (1 to 42 days), the broilers fed with a blend of citric and benzoic acid presented greater feed intake, without increment in weight gain. The data of allometry of the digestive system organs and the jejunal pH values were not influenced by the treatments. The citric acid increased the bacterial quantity of gram-positive coccus and total anaerobes in the jejunum. Under the conditions of the present study, we conclude that the citric and benzoic acids, isolated or combined, do not benefit the nutrition of broilers challenged with E. acervulina.(AU)


O objetivo deste trabalho foi avaliar os efeitos da inclusão isolada ou associada dos ácidos cítrico e benzoico na alimentação de frangos de corte sobre o desempenho, a alometria de órgãos do sistema digestório, o pH intestinal e a quantidade de microrganismos no jejuno. Foram utilizados 840 pintos de corte, machos, da linhagem Cobb, distribuídos num delineamento inteiramente ao acaso, em esquema fatorial 2 × 2, com suplementação ou não de ácido cítrico e suplementação ou não de benzoato de sódio, com sete repetições. Aos 14 dias de idade, foi inoculado, via oral, 1mL de solução contendo 1 × 105 oocistos esporulados de Eimeria acervulina por ave. Não houve efeito dos ácidos sobre o desempenho dos frangos no período de um a 21 dias. No período total (um a 42 dias), os frangos alimentados com a mistura de ácidos cítrico e benzoico apresentaram maior consumo de ração, sem incremento no ganho de peso. Os dados de alometria dos órgãos do sistema digestório e os valores de pH do jejuno não foram influenciados pelos tratamentos. O ácido cítrico aumentou a quantidade de bactérias do gênero cocos Gram positivos e anaeróbios totais no jejuno. Nas condições do presente estudo, conclui-se que os ácidos cítrico e benzoico, isolados ou associados, não beneficiam a nutrição de frangos de corte desafiados com E. acervulina.(AU)

ABSTRACT

Objective: Biological agents are among the medicines with the highest revenue in the world market. Biosimilars are copies of biological products introduced into the market to offer clinical efficacy like the originator or reference product at lower prices. This study aimed to verify the characteristics and price differences between biological medicines registered and marketed in Brazil until the end of 2019. Methods: All records were collected by November 2019 on the website of the National Health Surveillance Agency (Anvisa). The list of the Chamber of Regulation of the Medicines Market (CMED) consulted for the price analysis, has an economic classificatory criterion with eight ranges. Categorization, according to the date/period of authorization for marketing, was also made. Results: At Anvisa site, there are 144 drugs present in 277 products distributed in three regulatory categories: new, biological, and similar. Approximately 73% of drugs have been approved in the past five years. Three classes represent 77.9% of all drugs-antineoplastics and immunomodulatory agents (38.6%), blood and blood organ forming (20.7%), and alimentary tract and metabolism (18.6%). Of the 178 products listed in the CMED, 26 (14.6%) have prices above 10,000 reais. Conclusion: The prices of original products, for most of the inputs, are lower than those of biosimilars, reversing the international logic.

ABSTRACT

This study aimed to evaluate the performance, apparent digestibility of diet, ingestive behavior which occurred in two moments, carcass traits, being evaluated constituent and non-carcass components, and also the effect the yeast culture could promote in the peripheral temperature of rumen, hull and body temperature. The diets consisted of a constant ratio of 50% forage (maize silage) and 50% concentrate. Thirty-six steers, ½ Angus Nelore, with average age of 11 months and average initial body weight of 339.5±10kg were used in the experiment. The inclusion of yeast culture promoted a higher daily dry matter intake (8.83 vs 9.35kg day-1) and, consequently, a better daily weight gain (1,143 vs. 1,325kg day-1) in the initial feedlot phase, with no difference in other periods. The apparent digestibility of the diet containing yeast culture was higher than the control diet (69.69 versus 68.32%, respectively), and its use did not interfere with the feeding behavior of the animals. Based on our findings, supplementation with yeast culture may bring positive results in the initial feedlot phase.(AU)


O objetivo deste estudo foi avaliar o desempenho; a digestibilidade aparente da dieta; o comportamento oral ingestivo, o qual ocorreu em dois momentos, as características de carcaça, sendo avaliados componentes integrantes e não integrantes da carcaça; bem como o efeito que a cultura de leveduras pudesse promover perante a temperatura periférica de rúmen, casco e temperatura corpórea, sendo aferida por meio da temperatura retal. As dietas foram constituídas em uma constante relação de 50% de volumoso (silagem de milho) e 50% de concentrado. Utilizaram-se no experimento 36 novilhos, ½ sangue Angus Nelore, com idade média de 11 meses e peso vivo médio inicial de 339,5 ± 10kg. O uso de cultura de leveduras promoveu maior consumo diário de matéria seca (8,83 contra 9,35 kg dia-1) e consequentemente melhor ganho de peso diário (1,143 contra 1,325kg dia-1) na fase inicial do confinamento, não havendo diferença nos demais períodos. A digestibilidade aparente da dieta que continha cultura de leveduras foi superior à da dieta controle (69,69 contra 68,32%, respectivamente), e seu uso não interferiu no comportamento ingestivo dos animais. Com base nos resultados do presente estudo, a suplementação com cultura de leveduras pode trazer resultados positivos na fase inicial de confinamento.(AU)

ABSTRACT

Avaliou-se a eficiência da administração de subdoses de eCG nos acupontos Bai Hui e Hou Hai em protocolos de sincronização de estro em cabras. Na primeira etapa, 57 cabras foram distribuídas aleatoriamente em quatro tratamentos: T1- 300UI de eCG intramuscular (IM); T2- 60UI de eCG no acuponto Hou Hai; T3- 60UI de eCG no acuponto Bai Hui e T4- 60UI de eCG IM; e na segunda etapa, 28 cabras foram distribuídas aleatoriamente em três tratamentos: T1- 300UI de eCG IM; T2- 30UI de eCG no acuponto Bai Hui e T3- 30UI de eCG IM. Ao final do tratamento hormonal, as cabras foram monitoradas para detecção do estro, realização das coberturas e avaliação do comportamento reprodutivo. Os dados foram submetidos à análise de normalidade, seguida dos testes estatísticos adequados para cada variável. Na primeira etapa experimental, obteve-se maior duração de estro nas cabras do T1 (P=0,009). Na segunda etapa experimental, obteve-se maior número de animais em estros no T1 (P=0,03). As demais variáveis para ambas as etapas não sofreram influência dos tratamentos (P>0,05), demonstrando que a administração de subdoses de eCG nos acupontos Bai Hui e Hou Hai foi eficiente para sincronizar o estro.(AU)


The efficiency of administration of subdoses of eCG in the Bai Hui and Hou Hai acupoints in oestrus synchronization protocols in goats was evaluated. In the first stage, 57 goats were randomly assigned to four treatments: T1- 300UI of intramuscular eCG (IM); T2- 60UI of eCG in acupoint Hou Hai; T3- 60UI of eCG in the Bai Hui acupoint and T4- 60UI of eCG IM; and in the second stage, 28 goats were randomly assigned to three treatments: T1-300UI of eCG IM; T2-30UI of eCG in the Bai Hui acupoint and T3- 30UI of eCG IM. At the end of the hormonal treatment the goats were monitored for estrus detection, and evaluation of reproductive behavior. The data were submitted to normality analysis, followed by appropriate statistical tests for each variable. In the first experimental stage, a longer duration of estrus in the T1 goats (P= 0.009) was obtained. In the second experimental stage, a greater number of animals were obtained in estrus at T1 (P= 0.03). The other variables for both experiments were not influenced by the treatments (P> 0.05), demonstrating that administration of eCG subdoses in the Bai Hui and Hou Hai acupoints was efficient to synchronize the estrus.(AU)

ABSTRACT

Bredemeyera floribunda roots are popularly used to treat snakebites in the semiarid region of Northeast Brazil, and previous studies indicate the anti-ophidian actions of triterpenoid saponins found in its roots. To assess B. floribunda root extract (BFRE) activity against the effects of Bothrops jararacussu venom (BjuV), antiphospholipasic, antiproteolytic, antihemorrhagic, antinecrotic, and anti-edematogenic activities were investigated in mice. Phytochemical analysis revealed the presence of saponins, flavonoids, and sugars, with rutin and saccharose being the major constituents of BFRE. Acute toxicity was determined and BFRE was nontoxic to mice. Phospholipase A2 and proteolytic activities induced by BjuV were inhibited in vitro by BFRE at all concentrations tested herein. BFRE (150 mg/kg) inhibited paw edema induced by BjuV (50 µg/animal), reducing total edema calculated by area under the curve, but carrageenan-induced paw edema was unchanged. Hemorrhagic and necrotizing actions of BjuV (50 µg/animal) were considerably decreased by BFRE treatment. Thus, BFRE blocked the toxic actions of B. jararacussu venom despite having no anti-inflammatory activity, which points to a direct inhibition of venom's toxins, as demonstrated in the in vitro assays. The larger amounts of rutin found in BFRE may play a role in this inhibition, since 3′,4′-OH flavonoids are known inhibitors of phospholipases A2.

ABSTRACT

O presente estudo teve como objetivo avaliar o efeito da inclusão de óleo de peixe associado ao ácido ascórbico no diluidor para criopreservação de sêmen caprino. Dois machos da raça Boer foram submetidos à coleta de sêmen pelo método de vagina artificial, sendo os ejaculados avaliados quanto aos aspectos físicos e morfológicos. Após avaliação, formou-se um pool, seguido do fracionamento em cinco grupos: G1 - diluidor citrato-gema e G2, G3, G4 e G5 - diluidor citrato-gema acrescido de 1,0; 2,0; 3,0 e 4,0% de óleo de peixe e 0,05% de ácido ascórbico, respectivamente. Após descongelamento, foram realizadas avaliações físicas do sêmen e os testes complementares de termorresistência lento (TTR), hiposmótico (HO), integridade acrossomal e compactação da cromatina espermática. Houve comportamento linear crescente (P<0,05) para motilidade pós-descongelamento. Não houve diferença (P>0,05) para vigor pós-descongelamento (2,00±0,24). No TTR não houve diferença (P>0,05) para motilidade e vigor espermáticos entre os tempos cinco e 180min, com médias inicial e final de 62,17±12,13 e 14,29±10,55 para motilidade e de 2,00±0,52 e 0,49±0,44 para vigor. Não houve diferença (P>0,05) para o HO, com porcentagem média de espermatozoides reativos de 23,5±5,96%. Houve comportamento linear crescente para acrossoma íntegro e decrescente para acrossoma irregular (P<0,05). Não houve diferença (P>0,05) na compactação da cromatina, com 97,06±1,17% de cromatina íntegra. A inclusão até 4% de óleo de peixe acrescido de ácido ascórbico no diluidor melhorou motilidade e integridade de acrossoma após a criopreservação.(AU)


The study aimed to evaluate the effect of fish oil inclusion associated with ascorbic acid in the thinner for goat semen cryopreservation. Two male Boers underwent semen collection through the artificial vagina method, ejaculates being then assessed for physical and morphological aspects. After evaluation, a pool was formed, followed by the split into five groups: G1 - yolk-citrate extender and G2, G3, G4 and G5 - yolk-citrate extender plus 1.0; 2.0; 3.0 and 4.0% fish oil and 0.05% ascorbic acid, respectively. After thawing, physical evaluations of semen were assessed and additional testing slow heat resistance (TTR), hiposmotic (HO), acrosome integrity and compression of sperm chromatin. There was linear increase (P<0.05) post-thaw motility. No difference was obtained for post-thaw vigor and there was no influence of the association of fish oil and ascorbic acid in TTR. Plasma membrane integrity, by hyposmotic test (HO), presented a mean of reactive spermatozoa of 23.5±5.96% (P>0.05). There was linear increase for intact acrosome and decreasing acrosome irregular (P<0.05). In the analysis of the chromatin compaction, approximately 3% of damages (P>0.05) were observed. The inclusion of 4% fish oil plus ascorbic acid in diluter improved motility and acrosome integrity after cryopreservation.(AU)

ABSTRACT

In spite of the many available protocols, the use of chemotherapy for the management of canine mast cell tumours (MCT) remains empirical, and there is lack of criteria for the choice of protocol and definition of patients who may benefit from treatment. The objective of this study was to evaluate the outcome of dogs with MCT after adjuvant chemotherapy according to the risk of recurrence or metastasis proposed on the literature. This prospective study included 89 followed up dogs with prognosis assesment including clinical, histological, immunohistochemical and genetic features of canine MCT. Patients were grouped according to risk of recurrence and metastasis and recommended treatment with lomustine followed by chlorambucil if considered at high-risk, or vinblastine followed by chlorambucil if a patient was at intermediate risk. Outcome was defined by disease-free interval (DFI) and overall survival (OS) estimated by Kaplan-Meier curve. Adjuvant lomustine was useful for control of canine MCT of high-risk of recurrence or metastasis, but only when sequentially associated to chlorambucil with a DFI of 686 days and not reached OS. There was no difference in outcome in the intermediate-risk group despite choosen treatment. Patients at intermediate-to-low risk may not require adjuvant treatments, even in the absence of free surgical margins.(AU)


Apesar dos inúmeros protocolos disponíveis, o uso da quimioterapia permanece empírico para o mastocitoma canino e faltam critérios para escolha do protocolo e da definição dos pacientes que poderiam se beneficiar do tratamento. O objetivo deste estudo foi avaliar o resultado de cães com mastocitoma após a quimioterapia adjuvante, de acordo com o risco de recorrência ou metástase proposto na literatura. Este estudo prospectivo incluiu 89 cães com acompanhamento clínico e avaliação prognóstica, incluindo características clínicas, histológicas, imuno-histoquímicas e genéticas dos mastocitomas. Os pacientes foram agrupados segundo o risco de recorrência ou metástase, sendo recomendado tratamento com lomustina seguida de clorambucila, se considerados sob alto risco, ou vimblastina seguida de clorambucila, se estivessem sob risco intermediário. O resultado final foi definido pelo intervalo livre de doença (ILD) e pela sobrevida global (SG), estimados pela curva de Kaplan-Meier. Na adjuvância, a lomustina foi útil no controle do mastocitoma canino de alto risco, mas apenas quando associada ao clorambucila, com um ILD de 686 dias, sem atingir a mediana para SG. Não houve diferença no grupo de risco intermediário, independentemente do tratamento escolhido. Pacientes de risco intermediário podem não necessitar de tratamentos adjuvantes, mesmo na ausência de margens cirúrgicas livres.(AU)

ABSTRACT

Feline Injection Site-Associated Sarcoma (FISS) is a neoplasm that implies in reduction of quality of life and overall survival in feline patients. A retrospective study of 13 cases of FISS was conducted to evaluate the efficacy of surgical treatment associated to chemotherapy with doxorubicin or carboplatin. Local recurrence occurred in all patients. Patients treated with surgery and chemotherapy presented a longer overall survival and disease-free interval when compared to those that solely received surgical treatment, although no statistical significance was observed (p= 0.3360 and 0.7506, respectively). Surgery remains as the main option for FISS treatment. Further prospective studies with larger samples are warranted to investigate the benefit of chemotherapy for this neoplasm.(AU)


O Sarcoma de Aplicação Felino (SAF) é uma neoplasia associada a redução na qualidade de vida e sobrevida global. O objetivo deste estudo foi avaliar a eficácia da quimioterapia associada à cirurgia no manejo do SAF. Estudo retrospectivo de 13 pacientes com SAF submetidos à cirurgia isolada ou associada a quimioterapia com carboplatina ou doxorrubicina. Recorrência local ocorreu em todos os pacientes. Pacientes tratados com cirurgia e quimioterapia apresentaram maior sobrevida global e intervalo livre de doença quando comparados àqueles que receberam apenas tratamento cirúrgico, mas não foi observada diferença estatística (p=0,3360 e 0,7506, respectivamente). A cirurgia continua sendo a principal opção para o tratamento do SAF. Estudos prospectivos são necessários para investigação do real benefício da quimioterapia para esta neoplasia.(AU)

ABSTRACT

Electrochemotherapy is a local anticancer treatment in which non-permeant chemotherapeutic drugs are associated with electric pulses of well-established parameters. The electric pulses cause pores to open on the plasma membrane and facilitate drug transport, enhancing cytotoxicity and reducing side effects. Assessment of electrochemotherapy effects on Ehrlich solid tumor development in this work aims to evaluate in vivo usage of the electroporator device developed by the Department of Electrical Engineering of Engineering School of UFMG. Therefore, 40 Swiss mice were inoculated with Ehrlich tumor cells, and developed the tumor in solid form. After 21 days, mice were subjected to specific treatment protocols (control, bleomycin, electric pulses and electrochemotherapy); 17 days later they were euthanized and the tumors collected for histopathology analysis. Electrochemotherapy induced discrete weight loss and an inflammatory response in the tumor, which was not seen on the other treatment groups. Bleomycin alone induced necrosis. Both groups showed lower cellular proliferation rates. From this study, it was concluded that the animals tolerated electrochemotherapy treatment under anesthesia and the electroporator device developed by the Engineering School of UFMG was adequate when used in an electrochemotherapy protocol.(AU)


Eletroquimioterapia é uma modalidade de tratamento local contra o câncer em que a administração de quimioterápicos não penetrantes à membrana plasmática é associada à aplicação de pulsos elétricos com parâmetros bem estabelecidos, que abrem poros na membrana plasmática e facilitam a entrada desses fármacos nas células, aumentando sua citotoxicidade e reduzindo efeitos colaterais. A avaliação dos efeitos da eletroquimioterapia sobre o desenvolvimento do tumor sólido de Ehrlich em camundongos Swiss neste trabalho teve como objetivo testar o uso in vivo do aparelho eletroporador desenvolvido pelo Departamento de Engenharia Elétrica da Escola de Engenharia da UFMG. Para tanto, foram utilizados 40 camundongos fêmeas da linhagem Swiss, nos quais foram inoculadas células de tumor de Ehrlich, para o desenvolvimento do tumor na forma sólida. Após 21 dias, os camundongos foram submetidos ao protocolo de tratamento específico (controle, bleomicina, pulsos elétricos e eletroquimioterapia); 17 dias depois foram eutanasiados e seus tumores coletados para análise histopatológica e imuno-histoquímica. A eletroquimioterapia induziu perda de peso discreta e uma resposta inflamatória no tumor que não foi observada nos outros grupos. O grupo bleomicina apresentou maior porcentagem de necrose. Ambos os grupos apresentaram menor índice de proliferação celular. Com este estudo, pode-se concluir que o tratamento sob anestesia foi bem tolerado pelos animais e que o aparelho eletroporador desenvolvido pela Escola de Engenharia da UFMG é adequado para utilização em um protocolo de eletroquimioterapia.(AU)

ABSTRACT

Após validação de dois diferentes métodos de mensuração de pressão arterial sistólica (PAS) não invasivos ou indiretos (oscilométrico e Doppler vascular) com o invasivo ou direto (cateterização arterial - padrão ouro) em 12 gatos hígidos anestesiados de mesma faixa etária, os métodos não invasivos foram avaliados e comparados em 24 gatos hígidos e conscientes, divididos em quatro grupos de idade. Em cada animal, procedeu-se à mensuração da pressão pelos métodos Doppler e oscilométrico. O método oscilométrico foi o primeiro a ser realizado, e, logo em seguida, o método Doppler. Os valores considerados foram obtidos das médias de sete medidas consecutivas para cada método, com intervalo de 30 segundos entre elas. A primeira medida e as medidas discrepantes (variação ˃ 20% PAS), obtidas com sinais óbvios de estresse e/ou com movimentação do animal, foram descartadas. Os métodos de mensuração oscilométrico e Doppler, embora tenham apresentado médias estatisticamente diferentes em todos os grupos, quando comparados em cada grupo, apresentaram uma correlação significativa, alta e positiva. Ou seja, sempre que for obtido um valor elevado por um método, o mesmo fato ocorrerá com o outro método e vice-versa. Clinicamente, a diferença observada não é relevante, uma vez que a diferença média dos valores obtida pelos dois métodos, em cada grupo, foi menor que 1,1%.(AU)


After two different validation systolic blood pressure measurement methods (PAS), not invasive or indirect (oscillometric and doppler vascular) with invasive or direct (arterial catheterization - Gold Standard) in 12 anesthetized healthy cats of the same age group, not invasive methods were evaluated and compared in 24 healthy conscious cats, divided into four age groups. In each animal, we proceeded to measure the pressure by Doppler and oscillometric methods. The oscillometric method was the first to be performed, and, shortly thereafter, the doppler method. The values considered were obtained from averages from seven consecutive measurements for each method with an interval of thirty seconds between them. The first measurement and disparate measurements (range ˃ 20% PAS) obtained with obvious signs of stress and / or animal movement were discarded. Methods of oscillometric and doppler measurement, although statistically different averages were shown in all groups when compared, each group showed a significant high positive correlation, meaning that when a high value is obtained by a method, the same occurs with the other method, and vice versa. Clinically, the observed difference is not relevant since the mean difference values obtained by the two methods, in each group was lower than 1.1%.(AU)

Pancreatic adenocarcinoma is important in oncology because of its high mortality rate. Deaths may be avoided if an early diagnosis could be achieved. Several types of tumors overexpress gastrin-releasing peptide receptors (GRPr), including pancreatic cancer cells. Thus, a radiolabeled peptide derivative of gastrin-releasing peptide (GRP) may be useful as a specific imaging probe. The purpose of the present study was to evaluate the feasibility of using99mTc-HYNIC-βAla-Bombesin(7-14)as an imaging probe for Capan-1 pancreatic adenocarcinoma. Xenographic pancreatic tumor was developed in nude mice and characterized by histopathological analysis. Biodistribution studies and scintigraphic images were carried out in tumor-bearing nude mice. The two methods showed higher uptake by pancreatic tumor when compared to muscle (used as control), and the tumor-to-muscle ratio indicated that99mTc-HYNIC-βAla-Bombesin(7-14)uptake was four-fold higher in tumor cells than in other tissues. Scintigraphic images also showed a clear signal at the tumor site. The present data indicate that99mTc-HYNIC-βAla-Bombesin(7-14)may be useful for the detection of pancreatic adenocarcinoma.

Cardiac diseases promote alterations in the autonomic control of the heart, leading to an increase in heart rate and, as a result, a decrease in heart rate variability (HRV).The aim of this study was to evaluate if the development of heart failure secondary to degenerative mitral valve disease (DMVD) concurs with changes in autonomic modulation of heart rhythm which are assessed by long electrocardiography examination (Holter). Dogs were evaluated by clinical examination and echocardiography in order to be categorized into the following groups: Control (healthy; n=6), DMVD (disease without heart failure; n=8), and DMVD heart failure (disease with heart failure; n=13). Arrhythmias and frequency domain HRV were determined by Holter. Diseased animals, when compared to healthy, had significantly lower total power, which indicates overall HRV. DMVD heart failure dogs also showed other disturbances such as high incidence of supraventricular arrhythmias, high heart rate, little amount of pauses (2.0s long between consecutive heartbeats), longer time in tachycardia, shorter time in bradycardia, low high frequency (parasympathetic control), and high low frequency (sympathetic and parasympathetic control) when compared to control (p<0.05). As cardiopatias cursam com alterações do controle autonômico do coração, resultando em taquicardia e consequente diminuição na variabilidade da frequência cardíaca (VFC). O objetivo deste estudo foi avaliar se o desenvolvimento de insuficiência cardíaca secundária à degeneração valvar mitral (DVM) leva a alterações no controle autonômico do coração, as quais podem ser determinadas pela eletrocardiografia contínua (Holter). Cães foram distribuídos em grupos experimentais após avaliação clínica e ecocardiográfica da seguinte maneira: controle (saudáveis; n=6), DVM sem insuficiência cardíaca (n=8) e DVM com insuficiência cardíaca (n=13). Arritmias e VFC foram determinadas pelo Holter. Animais portadores de DVM, quando comparados ao controle, apresentaram diminuição significativa da potência total, a qual é representativa de toda a VFC. Somente cães doentes e com insuficiência cardíaca apresentaram incidência elevada de arritmias supraventriculares, frequência cardíaca aumentada, pequena quantidade de pausas superiores a 2,0s entre batimentos consecutivos, permanência por mais tempo em taquicardia do que em bradicardia, diminuído índice de alta frequência (indicativo de controle parassimpático) e elevado índice de baixa frequência (indicativo de controle simpático e parassimpático), quando comparados ao controle (p<0,05). ABSTRACT

We determined the inhibitory allelopathic effects of the volatile extracts of Cinnamomum zeylanicum Ness, Lippia sidoides Cham. and Cymbopogum nardus L. on seed germination and root growth of seedlings of Bidens pilosa. The experiment was conducted at the Seed Analysis Laboratory of the Department of Plant Science, Federal University of Ceará. For this end, we used oils at the concentrations of 0.01, 0.02, 0.04 and 0.08% (v/v). Five treatments were used for each of the oils arranged in a completely randomized design with four replications of 25 seeds. The seeds were sown in Petri dishes lined with filter paper moistened with distilled water and, aiming at the indirect contact with each oil, two sheets of filter paper were placed on top of the lid, in which three (3) mL of each oil solution were added. Then, the dishes were incubated in a germination chamber at 25°C. The pH did not contribute to alter the results; the volatile extracts of essential oils of C. zeylanicum, L. sidoides and C. nardus inhibited seed germination and root growth of seedlings of B. pilosa, which shows allelopathic potential; and the concentration of 0.08% of oils caused the overall deterioration of the roots and death of seedlings of B. pilosa.


Determinou-se o efeito alelopático inibitório dos extratos voláteis de Cinnamomum zeylanicum Ness, Lippia sidoides Cham. e Cymbopogum nardus L., sobre a germinação de sementes e o crescimento de raiz de plântulas de Bidens pilosa. O experimento foi conduzido no Laboratório de Análises de Sementes do Departamento de Fitotecnia da Universidade Federal do Ceará. Para isso, utilizaram-se óleos essenciais nas concentrações 0,01, 0,02, 0,04 e 0,08%. (v/v). Foram utilizados cinco tratamentos para cada óleo dispostos em delineamento inteiramente casualizado com quatro repetições de 25 sementes. As sementes foram semeadas em placas de Petri forradas com papel de filtro umedecido com água destilada e, visando o contato indireto com cada óleo, foram colocadas duas folhas de papel de filtro na parte superior interna da tampa, onde foi adicionado 3(três) mL da solução de cada óleo. Em seguida, as placas foram incubadas em câmara de germinação a 25ºC. O pH não contribuiu para alterar os resultados; os extratos voláteis de óleos essenciais de C. zeylanicum, L. sidoides and C. nardus inibiram a germinação de sementes e o crescimento da raiz de plântulas de B. pilosa, evidenciando potencial alelopático; a concentração de 0,08% dos óleos causaram a deterioração total das raízes e morte das plântulas de B. pilosa.

ABSTRACT

This work aims to determine the most suitable nonlinear model to describe the growth of female collared peccaries (Pecari tajacu). The monthly records of the weight of 10 captive female collared peccaries over a period of two years in the Brazilian Amazon Region were used. The growth models used were the Von Bertalanffy, Brody, Gompertz and Logistic. The parameters were estimated by using the NLIN procedure from the SAS application. The criteria used to verify the adjustment of the models were: asymptotic standard deviation (ASD); coefficient of determination (R²); average absolute residual deviation (ARD) and the asymptotic rate (AR). The Brody model and the Logistic model estimated the highest (19.44kg) and the lowest (19.18kg) asymptotic weight (A), indicating the lowest (0.0070kg/day) and the highest (0.0121kg/day) maturation rate (K). These results and the coefficients of phenotypic correlation that varied from -0.75 and -0.47 confirmed the antagonistic nature between these parameters. The Brody model estimated the lower value for ARD, a limiting factor for describing the lowest value for AR through this model. The Brody model showed the best adjustment for AR, although the other models also showed a suitable adjustment to the weight data of said species/gender. Based on the AR obtained in this work, the Brody model is recommended for adjusting the growth curve of the female collared peccaries. Depending on the estimated values, especially for K, this trait can be included in a selection index.


Com o objetivo de ajustar modelos não-lineares, foram utilizados registros mensais do peso de 10 fêmeas de cateto (Pecari tajacu) coletados durante dois anos, no criatório do campo experimental Álvaro Adolfo da Embrapa Amazônia Oriental, Belém, PA. Utilizaram-se os modelos de Von Bertalanffy, Brody, Gompertz e Logístico. Os parâmetros foram estimados usando o procedimento NLIN do aplicativo SAS. Os critérios utilizados para verificar o ajuste dos modelos foram: desvio padrão assintótico (ASD); coeficiente de determinação (R²); desvio médio absoluto dos resíduos (ARD) e o índice assintótico (AR). Os modelos Brody e Logístico estimaram, respectivamente, o maior (19,44kg) e o menor (19,18kg) peso assintótico (A), caracterizando a menor (0,0064kg/dia) e a maior (0,0113kg/dia) taxa de maturação (K), haja vista a natureza antagônica entre estes parâmetros, comprovada pela correlação fenotípica variando entre -0,75 à -0,47. O modelo Brody estimou o menor valor para o ARD, fator limitante para caracterizar o menor valor para o AR por este modelo. Considerando o AR, o modelo Brody apresentou o melhor ajuste, contudo, pelos valores encontrados, os demais modelos também apresentaram ajuste adequando aos dados ponderais da referida espécie/sexo. Com base no AR adotado neste trabalho, recomenda-se o modelo Brody para ajustar a curva de crescimento de fêmeas de cateto (Pecari tajacu). Em razão dos valores estimados, sobretudo, para a K, essa característica pode ser incluída em um índice de seleção. Contudo, estudos com grupos mais representativos e criados em outras condições se faz oportuno.

Avaliou-se o efeito da suplementação com geleia real sobre a morfometria do aparelho genital, resposta superovulatória e qualidade embrionária de coelhas. Trinta e seis fêmeas foram distribuídas em quatro grupos (G), sendo: G1 (n=9) formado por animais não suplementados com geleia real, e G2, G3 e G4 (n=9 em cada grupo) por animais suplementados com 10, 20 e 40mg/dia de geleia real. A superovulação consistiu na aplicação de 40UI de gonadotrofina coriônica equina, seguida por 40UI de gonadotrofina coriônica humana, via intramuscular, 48 horas após, e submetidas à cobrição natural. Os animais foram sacrificados, e os embriões coletados 72 horas após a cópula. Não houve diferença estatística entre tratamentos para as variáveis analisadas. O peso médio do aparelho genital foi de 10,88±0,38g; dos ovários - direito e esquerdo -, 0,28±0,02g; e o índice gonadossomático, 0,02±0,0g. O número médio de estruturas totais recuperadas foi de 9,2±1,4; de embriões viáveis, 8,7±1,4; e de degenerados, 0,5±0,2. Dos embriões viáveis, 5,6±0,8 foram classificados como grau I; 2,3±0,5, como grau II; e 0,8±0,2, como grau III. A suplementação com geleia real na dose de até 40mg/dia não apresentou efeito estimulador sobre o aparelho genital e a qualidade embrionária de coelhas.


The purpose of this paper was to assess the effect of supplementation of royal jelly on the morphometry of the genital tract, superovulatory response and embryonic quality in adult rabbits. 36 rabbits were distributed in four groups (G), as follows: G1 (n=9): no supplementation with royal jelly, G2, 3 and 4 (n=9): supplementation with 10, 20 and 40mg/day of royal jelly. The superovulatory protocol consisted in the application of a dose of 40UI of Equine Chorionic Gonadotrophin, followed by a dose of UI of Human Chorionic Gonadotrophin, intramuscularly, 48 hours later, and submitted to natural cover. The animals were euthanasiated and the embryos collected 72 hours after copulation. There was no statistical difference for the analyzed variables (P>0.05). The average weight for the female genital tract was 10.89±0.38g; the average weight of right and left ovaries was 0.28±0.02g and the average gonadosomatic index rate was 0.02±0.0g. The number of total recovered structures was 9.2±1.4. The average number of viable embryos was 8.7±1.4 and of degenerated embryos was 0.5±0.2. Viable embryos were morphologically classified as degree I: 5.6±0.8; degree II: 2.3±0.5; degree III: 0.8±0.2. Royal jelly did not present stimulatory effect on organs related to the reproduction of rabbits, nor was it effective to improve their embryonic quality at dosages of up to 40mg of in natura royal jelly.

ABSTRACT

Introducción: Las Guías Alimentarias son un instrumento educativo que contiene doce recomendaciones orientadas a promover dietas y estilos de vidas saludables para prevenir enfermedades crónicas de la población. Objetivo: Comparar el nivel de conocimientos y hábitos alimentarios pre y post capacitación en las Guías Alimentarias del Paraguay para niños mayores a 2 años, en familias de zona urbana y rural. Materiales y Métodos: Estudio Semi-experimental de Series Temporales realizado en jefes de familias, donde se estudiaron datos generales de la familia, datos socioeconómicos del hogar, educación alimentaria clasificada en conocimientos, actitudes y prácticas. Resultados: La muestra estuvo conformada por 32 adultos de zona urbana y rural, donde se compararon nivel de conocimientos antes y después de la intervención, se evidenció que ambas zonas aumentaron su conocimiento en forma similar. Respecto al conocimiento intra grupo, los jefes de familia aumentaron significativamente su nivel de conocimiento posterior a la capacitación. En relación a los hábitos alimentarios, 12 de las 14 variables consideradas sufrieron una modificación favorable tras la intervención. Los temas desarrollados durante la capacitación modificaron favorablemente los hábitos alimentarios de los encuestados. La frecuencia de consumo de panificados, azúcares y grasas disminuyó significativamente tras la capacitación. Conclusión: El impacto de la intervención fue positivo en ambas zonas al reflejar un aumento en el nivel de conocimientos y una modificación favorable en los hábitos alimentarios, de allí la importancia de fomentar la educación alimentaria-nutricional en la población. Introduction: Food Guides are educational instruments that contain twelve recommendations on how to promote diets and healthy life styles oriented to prevent chronic diseases in the population. Objective: To compare the level of knowledge and the eating habits before and after training families of urban and rural areas of Paraguay in Food Guides for children over the age of 2 years. Materials and Methods: semi-experimental Time Series study conducted in heads of families, where general information of the family, socioeconomic household data, and information on nutrition education in terms of knowledge, attitudes and practices were studied. Results: the sample consisted of 32 adults of urban and rural areas, where knowledge before and after the intervention was compared, and in which a similar increase of knowledge was evidenced. Regarding knowledge within the group, the heads of the families significantly increased their level of knowledge after the training. Regarding eating habits, 12 out of 14 variables suffered a favorable modification after the intervention. The topics discussed during the training favorably modified the eating habits of those who were surveyed. The frequency of consumption of bakery products, sugars and fats decreased significantly after the intervention. Conclusion: The impact of the intervention was positive in both rural and urban areas, as they reflected an increase in the level of knowledge and a favorable modification of eating habits, hence the importance of encouraging nutrition education among members of the population. ABSTRACT

Foeniculum vulgare Mill. é espécie herbácea da família Umbelliferae, nativa da região do Mediterrâneo e da Ásia Menor, amplamente distribuída em todo o território brasileiro. É conhecida popularmente como funcho ou erva-doce e usada na medicina como analgésico, digestivo, carminativo, diurético, expectorante, lactígeno, anti-inflamatório, e antiespasmódico. O extrato bruto etanólico para a verificação das atividades biológicas foi preparado a partir de sementes compradas no comércio. Para a realização do perfil fitoquímico foi utilizada a cromatografia em camada delgada analítica; a atividade antimicrobiana foi determinada pelo teste de difusão em disco de papel e da concentração inibitória mínima (CIM); a atividade antinociceptiva foi realizada pelo método de contorções abdominais em cobaias. Os micro-organismos testados foram isolados clínicos multirresistentes obtidos do Setor de Bacteriologia do Hospital das Clínicas de Pernambuco. O estudo fitoquímico identificou a maioria dos compostos secundários presentes na fração metanólica das sementes, sendo eles: triterpenos, glicosídeos de flavanóides, terpenos menores (monoterpenóides, sesquiterpenóide e diterpenóides), e açúcares redutores. Os resultados obtidos mostraram que o extrato etanólico apresentou maior atividade frente à Micrococcus spp. (CIM=250µg/mL). Os resultados da avaliação da atividade antinociceptiva demonstraram que apenas a dosagem de 298 mg/Kg quando comparado com o padrão indometacina conseguiu uma redução significativa no número de contorções abdominais dos animais. Estudos posteriores deverão ser realizados para a identificação e isolamento de alguns compostos secundários, bem como a realização de outros protocolos de analgesia.


Foeniculum vulgare Mill. (Umbelliferae) is a herbaceous plant of the family Umbelliferae, native to the Mediterranean and Asia Minor region and widely distributed all over the Brazilian territory. It is commonly known as fennel or "erva-doce" and has been used in medicine as analgesic, digestive, carminative, diuretic, expectorant, lactigenous, anti-inflammatory, and antispasmodic agent. Crude ethanolic extract for the verification of biological activities was prepared from seeds bought in the market. For the phytochemical profile, analytical thin-layer chromatography was used; the antimicrobial activity was determined by the paper disc diffusion test and minimum inhibitory concentration (MIC); the antinociceptive activity was verified by the method of abdominal writhings in the animals. The tested microorganisms were multiresistant clinical isolates obtained from the Bacteriology Sector of the Clinical Hospital of Pernambuco. The phytochemical study identified the majority of secondary compounds present in the methanolic fraction of seeds, as follows: triterpenes, flavanoid glycosides, smaller terpenes (monoterpenoids, sesquiterpenoids and diterpenoids) and reducing sugars. The obtained results showed that the ethanolic extract had greater activity against Micrococcus spp. (MIC=250µg/mL). Results of the evaluation of the antinociceptive activity demonstrated that only the dosage of 298 mg/Kg, compared to the indomethacin pattern, led to a significant reduction in the number of abdominal writhings in the animals. Further studies will be carried out for the identification and isolation of some secondary compounds, as well as other analgesic protocols.

Foi verificada pelo teste de ELISA indireto a resposta humoral contra os toxoides botulínicos C e D em bovinos de diferentes idades. O estudo envolveu 90 animais, que foram divididos em três grupos (n = 30), de acordo com a sua faixa etária; inferior a 2 anos de idade (G1), entre 2 e 5 anos (G2) e superior a 5 anos (G3). Os grupos experimentais foram vacinados com duas doses de vacina antibotulínica bivalente (C e D) comercial, nos dias 0 e 42 após a primo-vacinação (booster). Na avaliação, quando realizada 30 dias após o booster, os animais do G3 apresentaram maior produção de anticorpos (p < 0,05) em relação aos demais grupos. Entre o G1 e G2 não houve diferença significativa na resposta humoral contra a toxina C, no entanto, contra a toxina D, os animais do G1 apresentaram maior produção de anticorpos. Todos os grupos produziram uma resposta significativa de anticorpos contra as toxinas botulínicas após a 2ª dose da vacina bivalente comercial, principalmente contra o tipo D.


Humoral response of vaccinated cattle against toxins of clostridium botulinum types C and D at different ages. Cattle humoral response against type C and D botulinum toxoids (indirect ELISA) was verified in animals of different ages. The animals (n = 90) were divided in three groups (n = 30): group one (G1): less than two years old; group two (G2): from 2 to 5 years old; group three (G3): more than 5 years old. The groups were vaccinated with two doses [0 and 42 days after primary vaccination (booster)] of bivalent (C and D) antibotulinum vaccine. Group three had higher antibody production (p ≤ 0.05) compared to the other groups, 30 days after the booster. There was no difference (G1 and G2; p ≥ 0.05) in the humoral response against C toxin, however, against D toxin, group one had higher antibody production. It was possible to conclude that after two doses of the commercial bivalent vaccine all groups produced a significant antibody response against botulinum toxins, especially against D type.
